Replying to Kenya’s first innings 564, the Leewards finished on 718 for seven, the highest team score ever recorded in sponsored regional first-class cricket.
The 25-year-old Joseph scored 195, Tuckett stroked 142, his maiden first-class hundred, and Elsroy Powell contributed an unbeaten 61 for the Leewards to top Guyana’s 37-year-old team record of 641 for five against Barbados at Bourda.
From an overnight 470 for four, the Leewards batted throughout the morning session without any casualties as the Africans toiled in the batting-friendly conditions.
Joseph, playing a stodgy innings as he resumed at his overnight 143, pressed towards a double century and moved to within eight runs of the mark at the lunch break.
His 150 came in 502 minutes and with Tuckett on 63, the Leewards took lunch on 542 for four -- within 23 runs of first innings points.
Joseph had been dropped on 187 by wicketkeeper Maurice Ouma, who failed to clutch a bottom edge chance off pacer Lameck Onyango.
He departed soon after the restart, brilliantly caught by a diving Otieno at short extra cover as he attacked Onyango, ending a 153-run fifth-wicket partnership with Tuckett.
His 635-minute innings contained 15 fours and one six.
Wilden Cornwall faced only two balls and was caught behind off leg-break bowler Francis Otieno without scoring.
Tuckett, playing solidly, achieved his hundred before tea off 201 balls in 299 minutes with nine boundaries.
It was during the last session that the Leewards erased the regional team record, with Tuckett pushing on to get 142 and Number 8 batsman Elsroy Powell an unbeaten 61.
Tuckett belted 12 boundaries in his innings that ended when he lofted part-time seamer Ravindu Shah for Onyango -- running in from the square leg boundary and diving -- to clutch a spectacular catch.
Powell hit seven fours in his half-century knock, while Otieno (3-78) finished as Kenya’s top bowler.
Maurice Odumbe, who scored 207 in Kenya’s innings, was named man-of-the-match.
In round three this weekend, the Leewards tackle West Indies-B in Nevis and Kenya meet Trinidad and Tobago in Tobago.
