The tourists dismissed their hosts for 281 with both James Anderson and Matthew Hoggard claiming three wickets.
England then put on 50 runs in 11 overs before the close for the loss of Graham Thorpe, caught behind for 18, who had been promoted up the order.
However, England suffered a setback when Mark Butcher was carried from the pitch with a freak ankle injury.
The batsman went over on his ankle shortly before lunch and is now a doubt for the first Test.
Jamaica had resumed on 25-2, and England picked up three further wickets through Andrew Flintoff, Ashley Giles and Anderson.
Flintoff struck in the 11th over of the morning when a full-length ball bounced more than Danza Hyatt expected and Marcus Trescothick clung on to a sharp chance at first slip.
Giles then had Findlay caught by Nasser Hussain before Anderson got in on the act by having Lorenzo Ingram caught behind.
But they lost their way in the middle session of the day as the bowlers struggled to make inroads into an inexperienced Jamaican line-up.
Shawn Findlay and Mario Ventura both made half-centuries, but when Ventura was run-out for 53 after tea England finally began to find reward for their efforts.
Anderson looked a different bowler with the new ball and, with the third delivery with it, dismissed Evon McInnis, caught by wicketkeeper Chris Read, after an entertaining 41 from 27 balls.
Anderson and Read, making his fifth catch of the innings, combined again in the next over to dismiss Matthew Sinclair for 42 in the next over, before Hoggard wrapped up the innings to finish with figures of 3-47.
After the dismissal of Thorpe three overs from the close, Trescothick ended the day unbeaten on 21 with Paul Collingwood, Butcher's replacement, at the other end on six.
Simon Jones returned to training with the Test squad after his late arrival in Kingston on Monday evening.
The Glamorgan fast bowler, who delayed his trip to the West Indies to have scans on a sore shin, bowled for about 15 minutes in the nets at Sabina Park.
He is likely to feature in the next warm-up game against a Vice Chancellor's XI, which begins Friday, to prove his form for the first Test. (BBC Sport)
